摘要

Abstract

Objective To study the effects of indomethacin on the proliferation and invasion of colon cancer cell line HCT116 and its mechanisms. Methods The effects of indomethacin on the proliferation and invasion of colon cancer cell line HCT116 were measured by WST-8 method and TransweU chamber in vitro. The expressions of p-catenin, cyclin Dl, and matrix metalloproteinase-7 (MMP-7) protein were detected by western blotting method after indomethacin treatment. Results The proliferation of HCT116 was inhibited by indomethacin in a time and dose dependent manner, and the IC50 of 24 h, 48 h, and 72 h were 440.36 μmol/L, 323.9 μxmol/L, and 254.37 μmol/L, respectively; and the invasion of HCT116 was also inhibited by indomethacin in a dose dependent manner. The expressions of fi-catenin, cyclin Dl, and MMP7 were reduced after indomethacin treatment. Conclusions Indomethacin can inhibit the proliferation and invasion of colon cancer cell line HCT116, which may through the Wilt /(3-catenin signal pathway.
