Abstract
Objective To investigate the effect of matrine injection combined with cisplatin on telomerase activity and wtp53, hTERT mRNA expression of human hepatocarcinoma cell line SMMC27721. Methods The inhibitory rate was evaluated by using MTT method. The interaction between matrine and cisplatin was analyzed by means of Jin's formulation. Telomerase activity was analyzed by TRAP-ELISA. The expression of wtp53 and hTERT mRNA were detected by semi-quantitive RT-PCR. Results Matrine injection and cisplatin used alone could inhibit the proliferation of SMMC27721 cells, and there was simple addition or synergistic effect after treated with combined agents. Compared with single drug cisplatin group, the inhibitory rate was increased significantly (P ＜ 0.01), telomerase activity was decreased markedly (P ＜ 0.01), the expression of wtp53 mRNA was increased significantly (P ＜ 0.01) while hTERT mRNA was decreased markedly (P ＜ 0.01). Conclusion Matrine injection combined with cisplatin can obviously inhibit proliferation of SMMC27721 cells and display simple addition or synergistic effect, and its mechanism may be involved in inhibiting telomerase activity by up-regulating expressions of wtp53 and down-regulating expressions of hTERT gene.关键词
