脓毒症小鼠心肌P-选择素基因的表达及意义
Expression and significance of P-selectin gene in myocardium of septic mice
摘要
Abstract
Objective To investigate the expression and significance of P-selectin in myocardium of septic mice. Methods Cecal Hgation and puncture (CLP ) was used to establish the septic mice model. 72 female mice were randomly divided into 6 groups; control group (n = 12) , sham operation group (n = 12) , CLP 2 h group (n= 12), CLP 4 h group (n = 12), CLP 8 h group (n = 12), and CLP 12h group (n = 12). Mice in each group were sacrificed, and the blood sample and left ventricular myocardium were harvested. The expression of P-selectin mRNA was detect by reverse transcription polymerase chain reaction (RT-PCR), the activity of myeloperoxidase (MPO) in myocardium was detected by kit, and the serum level of cTnl was detect by enzyme-linked immunosorbent assay (ELISA). Results The expressions of P-selectin mRNA were significantly increased in CLP 2 h, 4 h, 8 h, and 12 h groups as compared with those in control group and sham operation group (P ＜ 0.05). The expressions of P-selectin mRNA in myocardium in CLP 2 h, 4 h, 8 h, and 12 h groups were positively correlated with MPO activity in myocardium and serum level of cTnl. Conclusion Sepsis could lead to the remarkable up-regulation of P-selectin mRNA expression in myocardium, which might play an important role in neutrophil infiltration and contribute to myocardial injury.关键词
