摘要

Abstract

Objective To investigate the characterization of gyrA gene mutation in fluoroquinolones-resistant Mycobacterium tuberculosis. Methods 60 Mycobacterium tuberculosis clinical strains, including 30 drug-resistance strains and 30 drug-susceptible strains, were identified and their drug-resistance in phenotype were tested by BACTEC-MGIT960, gyrA gene was amplified by PCR and then fragments were cloned into T-vector and sequenced. Results Among the 30 Mycobacteria tuberculosis drug-susceptible isolates, no sense mutation was identified in QRDR.19 mutant strains at 90,91 or 94 site of gyrA gene were identified in 30 drug-susceptible strains. There were 63.3% strains with gyrA mutation among 30 drug-resistant isolates in phenotype. Conclusion There is closely association between gyrA gene mutation and fluoroqunolones-resistant in phenotype for Mycobacterium tuberculosis.
