Induction of apoptosis by baicalein on human cervix cancer HeLa cells

Abstract
Objective: To investigate the effect of baicalein on cell apotosis of human cervix cancer HeLa cells and the possible mechanism. Methods: MTT assay was used to determine the cell growth inhibitory rate. Cellular apoptosis was detected by Hoechst 33258 fluorescent staining and Annexin-V/PI double-staining assay via flow cytometric analysis. The expressions of bcl-2,Bax,p53 and caspase 3 were determined by Western blot. Results; (1) After the treatment with different concentration of baicalein for 24 hours, HeLa cell growth was significantly inhibited (F<0.05). (2) The apoptosis rates of HeLa cells were significantly increased in response to baicalein treatment (P<0.01). ( 3 ) The expressions of Bax,p53 and caspase 3 were up-regulated (P<0. 05 to P<0. 01 ) ,but the expression of bcl-2 was down-regulated when HeLa cells were treated with baicalein for 24 hours. Conclusion: Baicalein can induce apoptosis of HeLa cells probably by regulating the expressions of gene assioated with apoptosis.关键词
