Abstract
Objective:To investigate the relationship between insulin-like growth factor I receptor and the cisplatin resistance in ovarian cancer cells SKOV3. Methods: The xenograft model in nude mouse with human ovarian cancer cells SKOV3 was established and tumor-derived cells were harvested:SKOV3-P(not treated) ,SKOV3-R(cDDP treated). The expressions of IGF-1 and IGF-1R in the SKOV3-P and SKOV3-R cells were detected by immunocytochem-istry (ICC ) and flow cytometry ( FCM). The inhibition effects of cDDP alone or combination with IGF-1 R inhibitor NVP-AEW541 on proliferation of the cells were measured by MTT assay. The apoptosis rate of the cells was detected by FCM. Results:The SKOV3-P and SKOV3-R cells expressed IGF-I and IGF-IR, supporting the existence of an autocrine loop, and the expression of IGF-1 and IGF-1 R in the SKOV3-R cells was significantly higher than that in the SKOV3-P cells(P<0. 05). NVP-AEW541 had anti-proliferative effect on the SKOV3-P and SKOV3-R cells in a dose-dependent and time-dependent manner. The inhibition rate of proliferation was significantly increased by cDDP in combination with NVP-AEW541. The apoptosis rate was increased by cDDP in combination with NVP-AEW541 (P<0. 05). The inhibition and apoptosis rates of the SKOV3-P cells were significantly higher than those of the SKOV3-R cells (P< 0.05). Conclusion: Inhibiting the insulin-like growth factor 1 receptor may revert the cisplatin resistance in ovarian cancer cells.关键词
