摘要

Abstract

Objective: To investigate the feasibility of differentiation of rabbits bone mesenchymal stem cells ( BMSCs) in to endometrial epithelial cells in simulate endometrial microenvironment in vitro. Methods: Rabbits BMSCs were isolated and identified, and endometrial conditioned medium was extracted. BMSCs were induced by endometrial conditioned medium and estrogen ( p-estradiol 1×l0-7mol/L) in vitro. Immunofluorescence and Western blot were used to test the expression of epithelial marker. Results: Rabbits BMSCs in vitro showed great proliferative potential and clonal growth. The positive rates of CD44 and CD90 in BMSCs were 99.91% and 98.64% respectively,and the expression of CD45 was negative. BMSCs morphology was differentiated from stromal cells to epithelioid cells by cultured in the endometrial conditioned medium and estrogen for 5 days. The epithelial marker cytokeratin was positive in immunofluorescence analysis in the differentiated BMSCs, and the expression levels of cytokeratin in the experimental group were higher than those in the control group through Western blot (P< 0. 05). Conclusion:Bone marrow mesenchymal stem cells have the potential to differentiate in to endometrial epithelial cells induced by endometrial conditioned medium and estrogen. The simulate endometrial microenvironment plays an important role in the process of MSCs differentiation.
