环氧化酶-2在大鼠动脉血栓中的表达及意义
Expression of COX-2 and its role in rat arterial thrombosis

Abstract
Objective Inflammatory chemokines are involved in the pathogenesis and prognosis of arterial thrombosis. This study was to investigate the expression of cyclooxygenase-2 (COX-2) in arterial thrombosis and its significance. Methods Rat arterial thrombosis model was induced by FeCl3 injury of the left carotid artery. Histological analysis of injured carotid arteries was performed by HE staining. Rats were sacrificed on day 1, day 3, day 7 and day 14 after FeCl3 injury, respectively. The expression of COX-2 protein in arterial thrombosis was assessed by immonohistochemistry (IHC). Expressions of COX-2 protein in arterial thrombosis and circulating white blood cells at different time were measured by Western blot assay. Results FeCl3 injury resulted in obvious mural thrombosis. IHC showed multiple sites of positive COX-2 staining in injured day 14 rats, while no positive staining was found in controls. Western blot analysis showed that COX-2 was expressed in minor degree in control group, but was high in the injured group, starting from day 1 and increased continuously to day 3, day 7 and day 14. The inflammatory cells in arterial thrombosis were neutrophils at day 1 and day 3, and were monocytic macrophage at day 7 and day 14. Conclusions COX-2 expression increases significantly in carotid artery thrombosis and the expression of COX-2 is related to arterial thrombosis, which indicates that COX-2 might be involved in the process of arterial thrombosis.关键词
