摘要

Abstract

Objective:To obtain the IFITM3 gene by human embryonic kidney cells 293 T induced with interferon rhIFN-α 2B in vitro and observe the IFITM3 gene expression in the transfected 293T. Methods: Human embryonic kidney cells 293T were induced with interferon rhIFN-α 2B in vitro. The complete cDNA of IFITM3 gene was amplified using RT-PCR; and the product was ligased to the pMD18-T vector. The recombinant plasmid pV-IFITM3 was constructed by subcloning the IFITM3 gene into the eukaryotic expression vector pVAX1 after sequencing. Research on expression and location of IFITM3 gene was conducted by tracfecting 293T cells. Results : The results shows that target gene could be expressed and had antigenic activity through RT -PCR and Western blot identification. The IFITM3 gene was mainly located in the outer membrane of 293 T cells observed by Laser scanning confocal microscope. Conclusion; The IFITM3 gene was obtained successfully and its expression was verified . This research provides valuable reference for the fur -ther study of IFITM3 related antiviral drugs and the mechanism of its anti -virus.
