摘要

Abstract

Objective: To study T cells lineage polymorphism of TCR Vα CDR3 in peripheral blood of ankylosing spondylitis (AS) patients so as to provide experimental basis to immune pathogenesis research in AS . Methods:T cells TCR Va 34 subfamilies in AS patients PBMC were amplified by RT -PCR method, then TCR Va CDR3 lineages polymorphism were analyzed by immunization scanning spectrum. Results:TCR Va CDR3 scanning spectrum of 10 AS patients all showed abnormal distribution peak , including monoclonal, oligoclonal/oligoclonal trend, skewing peak and irregular abnormal peak type subfamilies . A total of 17 subfamilies appeared that monoclonal proliferation of a single peak in 10 cases patients , the frequency in descending order were : Vαl9 subfamily that monoclonal proliferation of a single peak in three patients ; each Vα32, Va7 and Vαl. 2 showed in 2 patients with a single peak ; following showed a single peak. Conclusion: TCR Vα CDR3 lineage have significant polymorphism and spectrum drift characteristics in AS patients peripheral blood , which further indicate that T cells play an important role in immunological pathogenesis in AS . Mono clonal/oligoclonal expansion of T cells may be autoreactive T cells in the pathogenesis of AS , and this study provides a foundation for further research into the mechanism of AS.
