Abstract
Aim To investigate the influence of plumbagin on the proliferation of human hepatoma carcinoma cells HepG2 and SMMC-7721 and further research the mechanism of the influence. Methods HepG2 and SMMC-7721 were respectively co-cultured with plumbagin. Then, through microscopic imaging and MTT assay, the effect of plumbagin on the two kinds of cells was studied; finally, with real-time fluorescence quantitative PCR ( Qpcr ) analysis, the mechanism behind the effect was investigated. Results Both microscopic imaging and MTT assay analyses showed that plumbagin could significantly inhibit the proliferation of the two kinds of cells, the further Qpcr analysis showed that for the two kinds of cells, plumbagin could significantly up-regulate the Bax/Bcl-2 ratio ( P = 0. 0017 and P = 0. 0104 ) , while significantly down-regulate the transcriptional level of Cyclin Dl ( P = 0. 0287 and P = 0. 0165 ). Conclusion Plumbagin can significantly inhibit the proliferation of HepG2 and SMMC-7721 , and the inhibitory mechanism is related to the increase of Bax/Bcl-2 ratio and the decrease of the transcriptional level of CyclinDl.关键词
