Abstract
Objective To develop an HPLC-MS/MS method for the determination of azithromycin in human plasma in order to evaluate its bioequivalence. Methods After adding internal standard, methanol was adopted for precipitating protein. After centrifugation, HPLC-MS/MS was performed for analysis on the C18 column with the mobile phase of 10 mmol/L ammonium acetate buffer solution (containing 0.1% acetate acid) -methanol (35 : 65) and the selected fragment ions for detection. The healthy volunteers orally took the reference preparation and test preparation in a dose of 500 mg, respectively. After administration, the plasma azithromycin concentration was determined and the bioequivalence was calculated. Results The plasma azithromycin concentration in the range of 5. 91 -1 182 ng/mL demonstrated good linear relation. The lowest detection limit was 2. 37 ng/mL. Conclusion The established analytical method is sensitive, accurate, simple, convenient and suitable for the determination of plasma azithromycin.关键词
