脂联素启动子单核苷酸多态性频率及其水平与2型糖尿病的相关性
The study on the association of adiponectin promoter single nucleotide polymorphism frequencies and adiponectin levels with type 2 diabetes mellitus

Abstract
Objective To investigate the association of the levels of serum adiponectin( APN ) and the APN gene ( aPMl ) promoter region single nucleotide polymorphism ( SNP ) frequencies with the type 2 diabetes mellitus ( T2DM ). And it is expected to discover whether the aPMl promoter polymorphism is a genetic risk factor for T2DM. Methods Serum APN and biochemistry items were measured in the simple T2DM group, the T2DM with renal impairment group and the healthy control group. The serum APN levels were determined by ELISA. The correlation between the serum APN levels and other biochemical parameters was analyzed. And the aPMl polymorphisms were screened by the technique of denaturing high performance liquid chromatography ( DHPLC ). Results The serum APN levels of the healthy control group were significantly higher than that of the simple T2DM and the T2DM with renal impairment group ( P < 0. 01 ). And the serum APN levels were positively correlated with the high density lipoprotein ( P < 0. 01 ); negatively correlated with the fasting plasma glucose ( P < 0. 01 ); there was no significant correlation with cholesterol and triglyceride. SNP ( GG, GC, CC ) was presented at the promoter region of the aPMl-11377 sites. And the risk of suffering of T2DM in G allele carriers was significantly higher than the C allele carriers ( OR = 2. 09, P = 0. 028 ). Conclusion The APN may improve the insulin resistance. It may be the cardiovascular protection factor. And aPMl-11377C/G polymorphism is associated with the occurrence and the development of T2DM. It is indicated that the SNP-11377C/G polymorphism may increase the genetic risk of T2DM.关键词
