大黄ISSR-PCR反应体系的建立与优化
Establishment and Optimization of ISSR-PCR Reaction System for Rhubarb

Abstract
An orthogonal design combined with single factor experiment was applied to optimize ISSR-PCR amplification system of Rhubarb. The main elements of the reaction system ( Mg2+ 、 Taq DNA polymerase、dNTPs、 Primer、 DNA template) were tested . The result was analyzed by statistics software and direct analyzing . A suitable ISSR-PCR reaction system was established. Namely 25μL reaction system containing 30 ng DNA template, 2. 5μL 10 x Taq Buffer, 0. 4mmol/L dNTPs, 0. 25μmol/L primer, 2. 5 mmol/ L Mg2+ , 1. 25 U Taq DNA polymerase .关键词
