植物根际放线菌分离方法初探及根皮苷降解活性分析
Rhizosphere actinomycete isolation methods and phloridzin degradation activity analysis in plants

Abstract
[Objective]This research studied the effects of different medium and soil sample processing methods on plant rhizosphere actinomycete isolation and the mining of high activity phlorizin degradation strains with developmental potential to provide theoretical and practical guidance for the study on continuous cropping obstacle mechanism of apple and microbial remediation. [ Method ] From the 13 sampling provinces of Jilin , Hebei, Shandong , Shaanxi, Gansu , Henan, Hubei, Sichuan, Chongqing, Shanghai, Jiangxi, Anhui, and Zhejiang, 40 rhizosphere soil samples were collected. After naturally air-dried treatment, 10 mmol/L of MOPS solution and CaCO3 were used to pretreat the soil samples and then the actinomycetes were isolated in 7 different culture media by the spread plate method. The activities of isolated strains cultured in phlorizin selection medium were detected. [Result] The 7 kinds of culture media viz., B, MSC , MG, MT, Chi, HSG, and HV, respectively obtained 100, 78, 108, 64, 79, 113, and 88 strains of actinomycetes. The total isolated actinomycetes number obtained from MOPS solution treated soil samples was far greater than that obtained by the CaCO3 treatment. Out of all the 630 isolated strains, 29 strains were detected with phlorizin degradation activity. Also, out of all the isolated strains, 41 strains exhibiting high activity and morphological specific strains were analyzed by 16S rRNA gene sequencing, in which these strains were found belonging to 14 different genera of actinomycetes. [Conclusion] Out of the 7 different culture media, HSG culture medium displayed the best separation effect, MG displayed the second best separation effect, and MT displayed the worst separation effect. The soil treatment with MOPS solution was better than the CaCO3 pretreatment. Screening results showed that the rhizosphere actinomycetes strains contained some phlorizin degradation ability. The 16S rRNA gene sequencing results showed that the rhizosphere actinomycetes resource displayed an abundant biological diversity.关键词
