摘要

Abstract

Objective: To investigate the effects of hy- poxia and hypothermia on the expression of endothelin A receptor mRNA (ETA mRNA) and endothelin B receptor mRNA (ETB mRNA) in pulmonary tissue of newborn rats. Methods: Five to seven days old newborn rats were randomly divided into normal control (C) , hypoxia (H1) , hypothermal (H2) , and hypoxia/ hypothermal (H3) groups. Group C -was exposed to air at room temperature, group Hi to hypoxia (5%~ 6% O2) for 6 hours, group H2 to hypothermia (10 + l)℃ for 6 hours and group H3 to both of hypothermia (10 + 1) ℃ and hypoxia (5%~6% O2) for 6 hours. The changes of gross anatomy and histopathology (HE staining) in lungs -were studied. The expression of ETA mRNA and ETB mRNA -was measured -with FQ-RT-PCR. Results: (1) Expression of ETA mRNA increased significantly in groups H1, H2 and H3(H1, H2,P<0. 05; H3,P<0. 01). (2) Expression of ETB mRNA elevated significantly in groups H1, H2 and H3 (H1, H3 , P <0. 01 ; H2, P <0. 05). Conclusion: Over expression of ETA mRNA and ETB mRNA during pulmonary hemorrhage developing may play important roles by enhancing and attenuating ET-1 's damaging effects respectively.
