正交实验法优选血三七药材提取工艺
Optimum extraction process of Xuesanqi by orthogonal design
摘要
Abstract
To select the optimum extraction process of the roots of the Polygonum amplexi-caule var. sinense through orthogonal experiment. To consider the recovery of the dry extract and the the content of quercetin as complete index. Using L9 (34 ) orthogonal table screening, considering the ethanol concentration, extraction time, the capacity of ethanol and powder type as the main factors to choose the optimum extraction process. Quercetin was determined by HPLC. The mobile phase was methanol-0. 4% phosphorus acid glacial (49 : 51). The detection wavelength was set at 375 nm. The quercetin sample size showed a good liner relationship at the range of 1. 13~9. 04 μg (r=0. 9998), the optimal extraction technology was as follow: crushing the rhizome of Polygonum amplexicaule var. sinense into the coarse powder, extract it with the 90% ethanol twice six times' volume of 90% ethanol was added and extracted 1. 5h for the first, then four times' volume of 90% ethanol was added and extracted 1h.关键词
