摘要

Abstract

Mixed carbon sources culture medium with different ratio of glycerol to methanol was evaluated in the S-ade-nosyl-L-methionine ( SAM ) fermentative production by recombinant Pichia pastoris. When the supplement mixed medium contained 10% ( w/v ) glycerol, SAM production reached 6. 09 g/L, which was 20. 4% higher than the production of feeding 0% glycerol contained medium. On that basis, two sets of glycerol-methanol mixed-feeding strategy were performed at the 7 L fermentor to improve SAM production. When 100% methanol was fed into bioreactor for 24 h before 10% ( w/v ) glycerol content of mixed medium was put into use, SAM production was raised to 7. 94 g/L. When different glycerol content of mixed medium was fed into bioreactor at five different sections during the methanol induction phase, SAM production reached the highest 9. 80 g/L.
