石油污染土壤中降解菌的分离鉴定及降解基因筛选
Isolation, Identification and Gene Cloning of Oil-Degrading Bacteria in Contaminated Soil
摘要
Abstract
In order to get high degradation of oil bacteria, this study used oil as the only carbon source, chose samples from four places in the soil polluted by oil of Jinna oil field and purified the 14 strains of bacteria, 8 strains of actinomyces, and 9 strains of fungi. Through the physiological and biochemical reaction and identification of 16S rDNA, the C— 1 and H—1 strains were determined to belong to the genus bacillus. After degrading performance and optimization experiments, C—1 and H —1 growth curves were drawn and the best growth conditions and degradation rate were identified. Based on the design of 6 pairs of primers of degradation genes and the PCR method for cloning genes after referring to the relevant references, C—1 and H —1 bacteria were found out to contain genes of glutathione S—shift enzyme (GSTs) that can degrade biodegradable aromatic compounds.关键词
