Primary Culture of Rat Brain Microvascular Endothelial Cells

Abstract
Objective To develop a simple and reproductable method for the isolation and culture of brain microvascular endothelial cells (BMECs) of rats, and provide technical support for the studies of physiology, biochemistry and pharmacology of the brain endothelium and the studies of pathophysiology of cerebrovascular disease. Methods Relatively pure cortex explants were obtained froml-to 4-day-old neonatal Wistar rats by careful dissection, we used cortex explant to culture BMECs and observed the morphology and migration of endothelial cells from cortex explant. The cultured cells were identified according to the morphology and immunocytochemistry of factor VⅢ-associated antigen. Results We found that the cultured cells began to migrate from cortex explant after 24 hours, showed the spindle-shaped morphology and reached the monolayer confluence after 10-14 days. Immunocytochemistry demonstrated more than 95% of the third generation cultured cells were positive for VⅢ factor protein, which indicated the cultured cells were vascular endothelial cells. Conclusion Explant culture is a economic and easy method for primary culture of pure BMECs.关键词
