高产琥珀酸菌株的等离子体诱变选育
Screening of strain exposed under plasma discharge for improving succinic acid production
摘要
Abstract
The strain of Corynebacterium glutamicum was exposed on atmospheric and room temperature plasma ( ARTP) jet to improve succinic acid production. The physical parameters of plasma discharge and the optimum condition of mutagenesis were established. Adding the bromocresol purple as a pH indicator to the medium,the broth absorbances were determined to obtain the mutants having large change in light absorption by high-throughput screening at the wavelength of 590 nm. Results showed that when the survival rate of mutant strain was 8% , its positive mutation rate of Corynebacterium glutamicum cells was 22%. The results of the fed-batch culture in shake flask showed that the mutant strain E7 could produce succinic acid 15. 6 g/L and lactic acid 22. 8 g/L from 40 g/L glucose. The mutant strain could produce succinic acid and total organic acid.关键词
