Effects of epigallocatechin-3-gallate on the proliferation and apoptosis of human gastric cancer cell line MGC-803

Abstract
Objective To investigate the effect of epigallocatechin-3-gallate(EGCG)on apoptosis by acting on human gastric cancer MGC-803 cells and discuss its mechanism. Methods The cell proliferation inhibiting rate was determined by MTT method. After treated with EGCG, the apoptosis changes were observed by fluorescence microscope (AO/EB fluorescence stain) , DNA agarose gel electrophoresis and flow cytometry. The phosphorylation levels of mitogen-activated protein kinases (MAPK) , such as extracellular signal-regulated kinase (ERK) , c-jun N-terminal kinase (JNK) and p38, were examined by Western blotting. Results EGCG showed significant inhibitory effect on the proliferation of MGC-803 cells in a dose-and-time-dependent manners (all P < 0.05) , and typical features of the apoptosis were observed under AO/ EB fluorescent. When treated with 100, 200 μmol/L EGCG for 24 h, DNA extracted from MGC-803 cells displayed a characteristic ladder pattern on agarose gel electrophoresis. When treated with 50, 100, 200 μmol/L EGCG for 24 h, MGC-803 cells presented obvious subdiploid peaks. EGCG had an ability of inhibiting the activity of ERK and increasing the activities of p38 and JNK to some degree in MGC-803 cells. Conclusions EGCG can inhibit the proliferation of human gastric cancer MGC-803 cells in vitro and induce its apoptosis possibly through inhibiting the activity of ERK and increasing the activities of p38 and JNK in MGC-803 cells.关键词
