Effect on hepatic tissue of hepatic fibrosis in rats after local blockage of renin-angiptensin-aldosterone system at different levels

Abstract
Objective To observe the effects on hepatic histopathology of hepatic fibrosis in rats after local blockage of renin-angiptensin-aldosterone system at different levels. Methods Fifty SD rats aged 6 weeks were randomized into five groups, which were normal control group, model group, captopril group, losartan group and spironolactone group, with 10 rats in each. In the normal group, the rats received subcutaneous injection of olive oil subcutaneous injection of olive oil, while other rats were received subcutaneous injection of CC14-induced hepatic fibrosis. Next day, captopril group received intragastric instillation daily with captopril 60 mg/kg, losartan group received intragastric instillation daily with losartan 10 mg/kg, and spironolactone group received intragastric instillation daily with spironolactone 100 mg/kg. Control group and model group received normal saline everyday. Each group was sacrificed at 8 weeks. Hepatic fibrosis was evaluated by He-matoxylin-eosin staining and Masson staining. Results The degree of model group, captopril group, losartan group and spironolactone group hepatic fibrosis were higher than that in normal group(P <0.05). The degree of captopril group, losartan group and spironolactone group hepatic fibrosis were similar(P >0. 05) , and lower than that in the model group (P <0. 05) . There were no significant differences among the three treatment groups. Conclusion The blockage of renin-angiptensin-aldosterone system at different sites have the same inhibition of hepatic fibrosis.关键词
