单纯连续性小剂量糖皮质激素骨质疏松兔模型的建立
Establish the single-steroid-induced osteoporosis model of rabbit

Abstract
Objective To study the rabbit model of osteoporosis with the simple continuity of low-dose glucocorticoids model. Methods Forty-two female rabbits of 6-month age were divided into trail group and control group randomly. The rabbits of trail group were intragluteally subjected to 2 mg/ ( kg · d) methylprednisolone administration for 4 weeks, while the rabbits of control group were injected with equal normal saline. Some animals were sacrificed at 0, 2, 4 and 6 weeks. Then calcium, phosphorus, total cholesterol (TC) and triglyceride (TG) concentration were tested. Bone mineral density was measured with dual-energy X-ray absorptiometry. The micro-CT and HE staining were used to analyse bone mineral density and trabecular bone parameters followed by image processing and statistical analysis. And to analyse the biomechan-ical properties of the femur, tibia, lumbar spine. Results The rabbit body weight was significantly decreased after administration at 4 weeks. Compared with the control group, TC, TG had significant difference 2 weeks after administration ( P < 0.05 ) , calcium, phosphorus had significant difference until 4 weeks (P < 0. 05) ; BMD of femur and trabecular parameters were significantly decreased at 2 weeks ( P < 0.05) , significant difference of BMD and trabecular parameters were detected at 4 weeks (P <0.01); the BMD loss of Femur at 4 weeks to reach the basis value of 2.5 standard deviation. Conclusions It can establish glucocorticoid osteoporosis model by continuous administration a small dose of glucocorticoids. Not only proved hormone could induce osteoporosis, but also confirmed that a low dose of the hormone model can be established.关键词
