H2S对急性肺损伤大鼠PMN功能的影响机制

Abstract
Objective To observe the effect of hydrogen sulfide (H2S) on polymorphonuclear (PMN) function in lipopolysac-charide (LPS)-induced acute lung injury (ALI) rats. Methods 112 Sprague-Dawley rats were randomly divided into four groups:sham-operated group,model group(LPS were instilled intratracheally to induce ALI), LPS+NaHS (H2S donor) group,and LPS+PPG (propargylglycin, CSE inhibitor) group. Then according to the dosage time divided into two subgroups (group 4h and group 8h). All the rats were observed at 4 h (14 rats) or 8 h (14 rats)after agent administration. The blood PMN CDllb expression was detected by Flow cytometry. Seven rats of each subgroups,Lung tissue myeloperoxidase (MPO) activity was used to detect by Neighbors even fennel amine method. ICAM-1 protein content in lung tissues was calculated to measure by double antibody sandwich elisa method. Lung histomorphological changes were observed under microscope and index of quantitative assessment (IQA) was calculated to measure the lung injury severity. Seven rats of each subgroups were received bronchoalveolar lavage. PMN number in Bronchoalveolar lavage fluid (BALF) was counted under light microscope. Results Compared with sham-operated group,severe injuries of lung tissues and a raised IQA,a raised PMN number in BALF,expression of ICAM-1 protein and expression of MPO protein were observed in rats treated with LPS. Pre-administration of NaHS not only alleviated the injuries induced by LPS but also decreased IQA,PMN number in BALF,expression of ICAM-1 protein and expression of MPO protein. Pre-administration of PPG exacerbated the injuries induced by LPS. Also,increased IQA,PMN number in BALF,expression of ICAM-1 protein and expression of MPO protein were observed. Correlation analysis results showed that there was positive correlation between MPO activity in lung tissues and PMN number in BALF of rats ( r=0. 789, P<0. 01). Conclusion H2S can inhibit PMN function of ALI rats by reducing ICAM-1 ,CDllb expression and MPO activity.关键词
