Abstract

Objective To synthesize the probe of survivin antisense oligonucleotide modified gold nanoparticles by making use of the biological and optical characteristics of gold nanoparticles and to analyze its impact on A375 cells. Methods Citrate-stabilized gold nanoparticles were prepared using published procedures. Thiol-modified oligonucleotides were added to gold colloids to make nano-probes. Then the complementary reporter sequence was combined to prepare "nano-flare" probes. A375 cells were grown with media containing nano-flares probes. Fluorescent images were observed under the fluorescence microscope. Cell proliferation was measured by MTT and cell apoptosis was detected by Annexin V/PI double staining method. Results The nano-probes could inhibit A375 cell proliferation significantly (P<0.05) and induce cell apoptosis. Conclusion The nano-probes that can be stably synthesized can inhibit proliferation of A375 cells and induce their apoptosis.
