抗菌肽PR39在大肠杆菌中的表达及抑菌活性检测
Biological characteristics of the antibacterial peptide PR39 expressed in Escherichia coli

Abstract
According to the antibacterial peptide PR39 gene sequence in GenBank, a pair of primers was designed and synthesized. The PR39 gene segment was cloned to the express vector pCold, and thus the recombinant plasmid pCold-PR39 was constructed. Then the recom-binant plasmid was transferred into Escherichia coli BL21 (DE3). The analysis of SDS-PAGE and Western blotting showed that after induced with isopropyl β-D-1-thiogalactopyranoside (IPTG) , the target fusion protein was successfully expressed and identified in inclusion bodies. The protein had a molecular weight of 54 ku. The expressed PR39 protein showed obvious antibacterial activity. The in vitro antibacterial test showed that the maximum diameters of inhibition zones of recombinant antibacterial peptide PR39 against E. coli CVCC10141 and Salmonella typhimurium CVCC79102 were 5.7 mm and 4.9 mm, respectively. There was no antibacterial activity against Staphylococcus aureus CVCC25923. This study provides a basis for the application of PR39.关键词
