降糖调脂灵对2型糖尿病大血管病变的保护作用研究

摘要

Abstract

Objective To study the protective effect of Jiangtangtiaozhiling in T2DM angiopathy and to investigate possible mechanism. Methods 40 healthy Wistar rats were randomly divided into control group, model group, metformin group and TCM group with each group 10 rats. The control group was given standard diet, while other groups were given high glucose and fat diet + intraperitoneal injection of STZ to induce diabetes. The metformin group was given intragastric administration of 134 mg ? kg-1 · d-1 metformin and the TCM group was given intragastric administration of 690 mg ·kg-1 · d-1 Jiangtangtiaozhiling. The control group and model group were both given intragastric administration of 0. 9% sodium chloride injection in the same volume. After three months treatment, blood glucose, HbA1c, insulin, blood lipids ( TC, TG, HDL and LDL) of each group were detected and ISI was calculated. ELISA method was used to detect sTfR , RT - PCR method was used to detect TfR mRNA expression and transmission electron microscope was used to observe the cell infrastructure changes of artery smooth muscle. Results ( 1 ) The blood glucose, HbA1c, insulin, ISI, blood lipids, sTfR and TfR mRNA expression in artery smooth muscle in the four groups showed statistically significant differences ( P <0. 05 ) . The blood glucose, HbA1c, insulin, TC, TG, LDL, sTfR and TfR inRNA expression in the model group were significantly higher than in the control group, but the ISI and HDL were significantly lower than the control group ( P < 0. 05 ) . The insulin, bolld lipods, sTffl and it's inHNA between the metformin group and model group showed no statistically significant difference ( P > 0. 05 ) .The blood glucose, HbA,,, insulin, TC, TC, LDL, sTffl and TfR inHNA expression in the TCM group were significantly lower than in the model group, but the ISI and HDL were significantly higher than the model group ( P < 0. 05 ) . ( 2 ) Electron microscope observation showed that there were retro-grade changes in smooth muscle cells in the rats' aorta wall in model group, and the electronic density and heterochromatin in nu-cleolus were increased. Most mitochondria were so greatly damaged that a lot oi vacuoles appeared, and there were some big vacu-oles in Golgi complex. The metformin group had similar changes. The smooth muscle cells of rats, aorta wall in the TCM group were slightly damaged, and there were retrograde changes in mitochondria, Golgi complex and rough endoplasmic reticulum in a few smooth muscle cells, and some vacuoles appeared in mitochondria and rough endoplasmic reticulum. Conclusion Jiangtang-tiaozhiling has protective effect in T2DM angiopathy, and the mechanism may be that it can decrease blood glucose and blood lip-ids, thus improving iron metabolism disorder.
