胎鼠神经干细胞移植对大鼠脊髓损伤后神经元特异性烯醇化酶和皮质体感诱发电位的影响研究
Effect of Neural Stem Cells Transplantation on Neuron Specific Enolase and Cortical Somatosensory Evoked Potentials of Rat after Spinal Cord Injury

Abstract
Objective To study the effect of neural stem cells ( NSCs ) transplantation on neuron specific enolase ( NSE ) and cortical somatosensory evoked potentials ( CSEP ) of rat after spinal cord injury ( SCI ) . Methods SD rats were randomly divided into Brdu + NSCs group, NSCs group, SCI group and sham group. SCI models were induced with electrocircuit controlled spinal cord injuring device. NSCs in logarithmic phase were labeled with Brdu. The NSCs were transplanted into the injured site after SCI. HE staining was used to observe the pathological changes of spinal cord tissues, CSEP was used to monitor the recovery of spinal cord and immunohistochemisty method was used to observe NSE and the survival and migration of NSCs labeled by Brdu. Results There were significant differences on the latency of CSEP - P wave at each time point in four groups ( P < 0. 01 ), and there was no significant difference between Sham group and SCI group ( P > 0. 05 ), but there were significant differences between Brdu + NSCs and NSCs group ( P < 0. 01 ) . Compared with Sham group, there were significant differences on the latency of CSEP -P wave at each time point in SCI, NSCs and Brdu + NSCs groups ( P <0. 01 ); Except the first day, there were significant differences in NSCs and Brdu + NSCs groups compared with SCI group ( P <0. 01 ); there was no significant difference between NSCs and Brdu + NSCs group ( P >0. 05 ) . There were significant differences on the grey level and im-munostaining positive cells of NSE at each time point in four groups ( P <0. 01 ) and there were no significant differences at each time point in Sham group ( P >0. 05 ), but there were significant differences among SCI, NSCs and Brdu + NSCs groups ( P < 0. 01 ), then there were significant differences in SCI, NSCs and Brdu + NSCs groups compared with Sham group ( P < 0. 01 ); Except the first day, there were significant differences in NSCs and Brdu + NSCs groups compared with SCI group and there was no significant difference at each time point between NSCs and Brdu + NSCs group ( P > 0. 05 ) . There was a negative correlation between the number and grey level of NSE positive neurons and the latency of CSEP - P wave ( r = - 0. 937, P < 0. 001; r = - 0. 956, P <0. 001 ). Conclusion The transplantation of NSCs can survive and migrate in the injured site of spinal cord, and can up - regulate obviously the expression of NSE and shorten the latency of CSEP - P wave, then promote the functional recovery of hind limb in rat after SCI.关键词
