Abstract

OBJECTIVE: To explore the effects of paclitaxel on gene and protein expression of lung cancer cell line molecular A549 Wnt/β-catenin signaling pathway. METHODS: β-catenin protein of A549 cell expression of 1 nmol·L-1 paclitaxel group and control group were determined by immunohistochemical method. RT-PCR method was used to detect dkk-3 (cancer suppressor gene) and c-myc (primary cancer gene) mRNA expression level of A549 cell in 0.1, 1 and 10 nmol·L-1 paclitaxel groups and control group for 48 h. RESULTS: Compared with control group, the expressions of β-catenin protein and c-myc mRNA were all down-regulated in 0.1, 1, 10 nmol·L-1 paclitaxel groups, while dkk-3 mRNA expression was up-regulated significantly (all P< 0.05) in concentration-dependant manner. CONCLUSION: Paclitaxel could block Wnt signaling pathway and reduce the expression of c-myc by down-regulating protein expression of β-catenin and inducing dkk-3 mRNA expression of lung cancer cell line A549.
