摘要

Abstract

OBJECTIVE: To study on how to purify lysozyme highly and efficiently from egg white by ion exchange chromatography. METHODS: The egg white was diluted and then precipitated by isoelectric precipitation to remove ovalbumin and ovomu-cin, etc. The supernant flowed into ethyloic-agarose gel for ion exchange chromatography. Samples of elution peak was collected and freeze-dried. SDS-PAGE and bacteriolytic turbidimetry were used to identify the purity and activity of the products, respectively. RESULTS: After isoelectric precipitation and one-step column chromatography, the purity of the products was over 95% ,the specific activity was 17 600 U/mg,and the yield achieved 4.2 mg (lysozyme) per ml (egg white). CONCLUSION:One-step cation column chromatography can be applied to acquire high-quality lysozyme efficiently from egg white.
