Abstract

OBJECTIVE: To study the content change for 5 active components in Rhei Radix et Rhizoma decoction pieces before and after the irradiation by 60CO-γ ray. METHODS: Rhei Radix et Rhizoma decoction pieces were collected after the irradiation by normal dose of 60CO-γ ray. The contents of 5 main active components were determined by HPLC method (aloe-emodin, em-odin, rhein, chrysophanol, physcion). The determination was performed on SPHERIGEL ODS C18(150 mm×4.6 mm,5 μm) with mobile phase consisted of methanol-0.1 % phosphoric acid (80:20, V/V). The detection wavelength was set at 254 nm. RESULTS: The content of emodin in Rhei Radix et Rhizoma decoction pieces was the highest; followed by physcion and chrysophanol; the contents of aloe-emodin was the lowest. After irradiation, the contents of each component were lower than before, especially physcion; while less that of aloe-emodin. After half year, the main active components of un-irradiated Rhei Radix et Rhizoma decoction pieces decreased to different extents; the contents of main components in Rhei Radix et Rhizoma decoction pieces decreased to some extents after irradiation by 60CO-γ ray, and stability of the components increased. CONCLUSION: The effects of ray on chemical components should be considered during medicinal material or preparation sterilization. The index of preparations should be re-detected and analyzed quantitatively after sterilization to guarantee the contents of active components within national standard.
