Abstract

Objective To observe the changes of molecular chaperone glucose-regulated protein 94 (GRP94) in the hippocampus of post-traumatic stress disorder rat model. Methods Totally 45 rats were randomly assigned to three groups including a normal control group (nd, n =15) and two SPS groups (sps-7d,n =15;sps-14d,n =15),and prepared for single-prolonged stress (SPS) model. Immunohistochem-istry, Western blot and reverse transcription-polymerase chain reaction (RT-PCR) were performed to examine the changes of GRP94 in the rat hippocampus. Results By using immunohistochemistry and Western blot,the expression of GRP94 in the group of sps-7d was markably increased compared with the normal control group (P < 0.05). The expression of GRP94 in sps-14d group showed a significantly decreased compared to the sps-7d group (P < 0.05). The mRNA expression of GRP94 in RT-PCR had the similar trends of changes. Conclusion SPS would induce changes of GRP94 expression in the rat hippocampus, which provided basis for future study of neuronal apoptosis induced by endoplasmic reticulum stress.
