姜黄素抗C57BL/6小鼠肺纤维化与CathepsinK相关性研究
Effect of Curcumin on Anti-lung Fibrosis of C57BL/6 Mice and Its Relationship with Cathepsink

Abstract
Objective ;To investigate the mechanism of curcumin on anti - lung fibrotic effect in bleomycin induced lung fibrosis C57BL/6 mice. Methods; Intratracheal application of bleomycin in C57BL/6 mice was done. Briefly, tracheas of anaesthetized mice were surgically exposed and 0. 025 U of bleomycin diluted in sterile saline or saline only was slowly instilled . The mice were divided into three groups at random; Control, Bleomycin, Bleomycin + Curcumin (200 mg/ ( kg ·d). From the second day after bleomycin instillation, mice were challenged with saline or curcumin (200mg/(kg ·d) ) in 0.5% carboxymethylcellulose via oral gavage and sacrificed at 7 ,14 and 28 days post - treatment followed by determination of lung fibrosis by estimating lung collagen content. In addition, changes in cathepsin K protein and mRNA levels in the lung were determined by immunohistochemistry, western blotting and real time RT - PCR. Results: Curcumin treated mice responded with a lower collagen deposition in their lungs, which was accompanied by a significantly higher lung resistance compared with bleomycin -treated mice. Moreover, Curcumin treated mice responded with a lower degree of lung fibrosis than only bleomycin - treated mice. In addition, Cathepsin K protein levels were strongly increased in curcumin treated mice relative to only bleomycin treated mice( 14d, P =0. 006;7d,P =0. 015;28d, P =0. 027); curcumin also obviously increased the level of Cathepsin K mRNA(7d, P =0.047;14d,P =0.001). Conclusion:Curcumin increases the expression of cathepsin K and reduces lung collagen deposition in response to bleomycin challenge, thereby providing at least partial protection against bleomycin - induced lung fibrosis.关键词
