绵羊PRNP密码子136和154及171等位基因型的ARMS-PCR鉴定方法的建立
Establishment of ARMS-PCR genotyping method for identification of ovine PRNP alleles at codon position 136,154 and 171

Abstract
The specific primers targeting on the ovine PRNP genotypes were designed referenced on the ovine PRNP alleles at codon 136,154 and 171 obtained by our laboratory and synthesized. In terms of the type and position of mismatched base at the 3'end,optimal primers were selected and the amplification conditions were optimized. Then, genotyping ARMS-PCR method was established. This method was used to analyse individual sheep blood samples with known PRNP genotype, and the accordance rate was verified by comparing ARMS-PCR with PCR-SSCP. As a result, 15 known PRNP alleles, from 94 blood samples were detected by ARMS-PCR method with the accordance rate of 100%. The genotyping ARMS-PCR method is simple,rapid and cheap for accurate identification of each PRNP allele of ovine at codon position 136,154 and 171,and it was suitable for investigation of ovine PRNP allele type.关键词
