Apoptosis effect of hymenocallis littoralis alkaloid on human hepatoma cell HepG-2
摘要
Abstract
This paper explored the apoptosis of human hepatocellular carcinoma HepG - 2 cells by hymenocallis speciosa alkaloids ( HLA). The growth inhibition of human hepatocellular carcinoma HepG - 2 cell was detected by MTT method. The effect of alkaloid hymenocallis littoralis on the HepG -2 morphology was observed with fluorescence microscopy. The apoptosis rate was tested by flow cytometry. MTT showed that hymenocallis littoralis alkaloid has cytotoxicity to the HepG - 2 cell. Under the fluorescence microscope, the apoptotic HepG - 2 nucleus which was stained with Hoechst33258 showed a dense strong fluorescence and apoptotic bodies. The apoptosis rate which was detected by propidium iodide (PI) single staining with flow cytometry increased with the increase of hymenocallis littoralis alkaloid after 72 h. According to the above experiment, it could be concluded that hymenocallis littoralis alkaloid could induce the apoptosis of human hepatoma cells HepG -2. It is provided an evidence for the research and development of new drugs.关键词
