NIRF Inhibit the Replication and Expression of Hepatitis B Virus in the HBV Mouse Model by Hydrodynamic Transfection Method

Abstract
With the process and development of recent research on NIRF (Np95/ICBP-90 like RING finger protein), it has involved in cellular cancerization on certain conditions and epigenetic regulation, but the effect of NIRF on the replication and expression of hepatitis B virus (HBV)m vitro is unclear now. Here, an animal model was established by hydrodynamic transfection of the plasmid pAAV-HBV1.3 in BALB/C mouse to explore the connection between NIRF and the replication and expression of HBV. The serum and liver tissue at different time point were collected. The virus load in serum and liver were assayed by real-time PCR. The expression of HBc (hepatitis B virus core) protein was assayed by Western blot. The secretion of HBeAg was determined by ELISA and HBsAg, HBcAg were determined by immunohistochemical staining. The viral load in serum and liver in experimental group which co-injected with pAAV-HBV1.3 and NIRF were significantly lower than that in control group (n=3, P<0.05), meanwhile, the expression of HBc and HBeAg, HBsAg, HBcAg were inhibited by NIRF, the replication and expression of hepatitis B virus in the HBV mouse model were extended by NIRF by hydrodynamic transfection method, which pave a way for a better understanding of the mechanisms of HBV and drug development against hepatitis B virus.关键词
