Abstract

Objective To explore the clinical value of directly detecting measles virus-specific nucleoprotein gene( MV-N )from clinically diagnosed measles patients' urine using reverse transcription-polymerase chain reaction( RT-PCR ). Methods Urine was collected from 15 patients clinically diagnosed with measles from 2010 to 2011 in Bengbu, Anhui province. RNA was extracted from urine. RT-PCR was used to amplify 515 bp nucleotide fragment of the measles virus nucleoprotein. Serum of the same patient was collected to detect MV-IgM antibody. The positive rate was compared between two methods. Results Positive products were successfully amplified in 14 of 15 patients by RT-PCR method. The positive rate was higher by RT-PCR method than by MV-IgM detection method( 93. 3% vs 60. 0%, x2 = 9. 60 ,P = 0. 002 ). Conclusion RT-PCR for detecting the urine can be used as an early fast-assisted diagnosis method for measles virus, especially for patients with early rash. In the meantime, re-examination of negative MV-IgM in serum could reduce the missed diagnosis.
