心衰大鼠β3AR自身抗体的产生与心肌重塑的关系
Study on the emergence of autoantibody against beta3 adrenoceptor and its relationship with cardiomyocyte remodeling in heart failure rats

Abstract
Objective To explore the relationship of autoantibody against β3 adrenoceptor and cardiomyocyte remodeling during the development of heart failure(HF). Methods The 10-week old healthy male Wistar rats were randomized into two groups ;HF group and sham group. Heart failure rat models were induced by constricting the abdominal aorta. The level of the autoantibody against beta3 adrenoceptor was detected by ELISA before and after treatment for 4,8,12,16 weeks,and beta3 adrenoceptor in cardiac cells was tested by Western blot. Results ①The heart failure model was successfully constructed as evidenced by multifocal degeneration of myocyte, deposition of collagen and impaired cardiac function after 8 weeks of abdominal aorta banding. ②During the development of heart failure, the content of cardiomyocyte β3AR increased significantly with the elapse of time and maintained at a high level until 16 weeks after treatment. ③The OD value of β3 adrenoceptor autoantibody in the sera of heart failure models were significantly increased after 4 weeks of treatment(P<0.05) .peaked at week 8(P<0.01) and maintained a high level until 12 weeks after treatment(P <0.05) , and then decreased 16 weeks after treatment (P > 0. 05). Conclusion These results suggest that the up-regulation of myocardial β3AR might be a crucial factor involving in the stimulation of genesis of β3AR autoantibody during the course of heart failure.关键词
