苯并(a)芘对体外培养小胶质细胞炎性因子和P38的影响
Effects of Benzo (a) pyrene on cytokines and P38 of BV-2 microglial cells in vitro
摘要
Abstract
Objective To explore the effect of benzo (a) pyrene [B(a)P] on the cytokines production and its relationship with P38 activation in BV-2 cells. Methods Cells at exponential growth phase in vitro were treated with different concentrations of B(a)P(0,1.0, 2.0,4.0,8.0,16.0 μmoL/L) or dimethyl sulfoxide( DMSO,solvent control)for 16 h respectively. The supernatants were collected and analyzed by EL1SA to quantify the release of inflammatory factors IL-1β from the cells. Western blot was used to measure the expression of P38. Results In BV-2 cells, IL-1β secretion increased in a dose-dependent manner after B(a)P( P ＜0. 05 ). The level of P38 increased in a dose-dependent manner after B(a)P. Conclusion B(a)P might activate the inflammatory factors IL-1β secretion by up-regulating P38 expression, which produce the toxic effects on the nervous system.关键词
