Abstract
Objective To investigate radiosensitization effect of valproic acid on human glioma carcinoma cell line SHG-44 and its mechanism. Methods SHG-44 cells were treated with 0.5, 1.0, 2.0, 4.0 and 8.0 mmol/L sodium valproate, respectively. The radio sensitization of valproic acid on SHG-44 cells was measured by colony formation assay. The proliferation inhibition rate of valproic acid on was investigated by MIT assay in different time and dose. The apoptosis and cycle of SHG-44 cells in different groups were analyzed by flow cytometry (FCM). Results After treated with valproic acid, the cell proliferation was inhibited significantly in a time-and dose-dependent manner. The valproic acid have radiosensitization on human glioma carcinoma cell line SHG-44, SER = 1.32. Compared with the control group, cell apoptosis and the proportion of G0/G1 phase were increased significantly in the combination group, and the proportion of S phase was reduced that assayed by FCM. Conclusion Valproic acid can increase the radiosensitization of SHG-44. Its role may be to induced cell apoptosis, change cell cycle phase distribution and other mechanisms.关键词
