Effects of endothelial progenitor cells and VEGF on neovascularization in proliferative diabetic retinopathy

Abstract
Objective To investigate the effects as well as the interaction between endothelial progenitor cells (EPCs) and vascular endothelial growth factor (VEGF) on the occurrence and development of proliferative diabetic retinopathy (PDR) by detecting the fibrovascular membranes in PDR patients. Methods We collected 10 PDR membranes stripped from vitreous surgery; 10 PVR membranes and 1 human retina served as controls. Morphology of the membranes was observed. VEGF expression and EPCs counting were observed by immunohistochemistry. Results PDR membranes consisted of the central avascular area, the middle fiber structure with a few cells, and the peripheral rich cell area. Cells were distributed disorderly in PVR membranes. EPCs were found in all the 10 PDR membranes (5.90±1.83) and in only 1 PVR membrane (0. 10±0. 32), but not in human retina. VEGF expression in PDR membranes was significantly higher than in the control group (P<0.001). The number of EPCs was positively correlated with VEGF expression in PDR membranes. Conclusion EPCs and VEGF participate in and have a synergistic effect on neovascularization in PDR.关键词
