假臭草ISSR-PCR反应体系的建立与优化
Establishment and Optimization of ISSR-PCR Reaction System for Eupatorium catarium
摘要
Abstract
The effects of 5 factors ( Mg2+ concentration, dNTPs concentration, Taq DNA polymerase dosage, primer concentration and DNA templates concentration) on ISSR amplification were tested, and the annealing temperature was selected and optimized for establishment of a suitable ISSR-PCR reaction system of Eupatorium catarium through single factor test and orthogonal experiment design of L16 (4°). The results showed that the optimal ISSR-PCR reaction system (20 μL) contained 2. 0 mmol/L Mg2+,60 ng DNA template, 1. 5 U Taq DNA polymerase,200 μmol/L dNTPs and 0. 3 jumol/L primer. The establishment of the ISSR-PCR reaction system could lay a favorable foundation for the analysis of the genetic diversity of E. Catarium and for the exploration of prevention and control strategy for E. Catarium.关键词
