Abstract

Objective To investigate the effect of salviae miltiorrhizae on hypoxic mouse cardiomyorates. Methods Mouse cardiomyocytes were isolated from neonatal mouse heart and the cells were divided into 3 groups: high dose salviae miltiorrhizae (300 μg · mL-1 ) group, low dose salviae miltiorrhizae (75 μg · mL-1 ) group and control group, respectively. Cell viability was assessed was also evaluated using CCK-8 assay. After the cells were cultured under hypoxic condition (5% CO2 and 95% N2) for 12 h, cell apoptosis was assessed by Annexin V-PI and Caspase-3 assay. Gene expression of Akt, Bad and FCF after cell harvest using real-time PCR. Results High dose salviae miltiorrhizae significantly increased the cell viability as compared with the other two groups (both P<0. 05). Low dose also increased cell viability significantly as compared with the control group (P<0.05). High dose salviae miltiorrhizae significantly reduced cell apoptosis detected by Annexin V-PI as compared with the low dose group and the control group ( both P<0. 05 ) . The Caspase-3 activities in both salviae miltiorrhizae groups were significantly decreased as compared with the control group ( both P<0. 05 ). There was no significant difference between high dose group and low dose group in Caspase-3 activity. Real-time PCR showed that high dose salviae miltiorrhizae treatment significantly upregulated the expression of Akt and FCF (P<0.05) and reduced the expression of BAD (P<0.05) as compared with the control group. The gene expression was significantly higer for FCF (P<0.05) and lower for BAD (P<0.05) in the high dose salviae miltiorrhizae group as compared with the low dose group. Conclusion Salviae miltiorrhizae treatment increases cardiomyocyte viability and decreases cell apoptosis, mainly through the elevated gene expression of Akt, FCF and decreased gene expression of BAD.
