Abstract
Objective:To investigate the variation of (Th) 17-related cytokines interleukin-17 (IL-17) and interleukin-23 (IL-23) in peripheral blood and live before and after therapy in newborn mice with MCMV hepatitis. Methods: The mice were infected by intraperitoneal (i. p. ) injection of Murine Cytomegalorirus( MCMV group) , the control group were injected the same amount of sterile saline by intraperitoneal, the treatment group were injected ganciclovir of 60 mg/ (kg ·d) the same day after inoculation MCMV daily for 2 weeks. Serum alanine aminotransferase( ALT) , IL-17, IL-23 protein from blood serum, IL-17mRNA, IL-23mRNA in the liver of mice were assessed by automatic biochemical analyzer, ELISA method and semi-RT-PCR respectively. Correlation analysis were done among IL-17, IL-23 and ALT. Results;IL-17, IL-23 protein levels in the serum of virus groups on the day 3,7,14 after infection were significantly increased than the normal control groups in the corresponding time (P < 0.01). On the day 7,14 after treatmented by ganciclovir the IL-17,IL-23 levels were significantly lower than the virus groups in the corresponding time (P<0.01). IL-17mRNA, IL-23 mRNA expression in the liver tissues of MCMV mice were significantly increased than the normal control groups on the day 3,7,14 after infection (P <0.01). On the day 7,14 after treatment the IL-17mRNA, IL-23mRNA expression in the liver tissues were significantly lower than the virus groups. And IL-17, IL-23 were positively correlated with ALT levels (r = 0.691 -0. 803 , P < 0. 05) . Conclusion :The IL-17 and IL-23 increased after MCMV infection, which suggested that IL-17 and IL-23 may involve in the pathogenesis of MCMV hepatitis.关键词
