Abstract

Objective To establish a method for determining the content of luteolin and apigenin in Platy-codon Grandiflorus by high-performance liquid chromatography (HPLC). Methods HPLC was performed on an Alltima C18 column (4. 6 mm × 250 mm, 5 μm) with a mobile phase of acetonitrile-0. 5% phosphoric acid (35 : 65) at a flow rate of 1. 0 ml/min, a detection wavelength of 340 nm, and a column temperature of 25 ℃. Results Luteolin and apigenin had a good linear relationship between concentration and peak area in the concentration ranges of 11. 20-67. 20 μg/ml (r=0. 999 6) and 12. 25-73. 50 μg/ml (r=0. 999 9) , respectively. The average recovery rates of luteolin and apigenin were 99. 03% (RSD = 0. 77% , n=6) and 99. 71% (RSD=1. 38%, n= 6), respectively. Conclusion This HPLC method can be used for determining the content of luteolin and apigenin in Platycodon Grandiflorus due to its simplicity, accuracy, and reliability.
