Abstract

Airn :To observe the effect of fibrinogen-like protein 2( fgl2 ) gene silencing on myocardial microvascular endothelial cells( MMVECs ) proliferation and cell migration and whether Ang-1 and Ang-2 involve or not. Methods : We constructed a lentiviral RNA interferenced vector of human fgl2 gene. MMVECs were transfected by lentiviral vector. Four days after transfection,quantitative real-time RT-PCR was used to detect the expressions of fgl2,Ang-1 and Ang-2 mRNA,MTT assay was used to detect cell proliferation,and Transwell was used to detect cell migration. Results:The expression of fgl2 mRNA was decreased significantly compared with control group and GFP group( F=180. 301 ,P <0. 001 ). While the expressions of Ang-f and Ang-2 mRNA was increased( F = 59. 450 and 54. 572, P < 0. 001 ). The proliferation and migration ability of MMVECs increased( F =5. 341 and 6. 122, P < 0. 05 ). Conclusion : Fgl2 gene silencing induces MMVECs proliferation and cell migration. The possible mechanism is that fgl2 gene silencing upregulates the expressions of Ang-1 and Ang-2 mRNA.
