Protective function of reducing glutathione on airway epithelial cells in guinea pig models of asthma

Abstract
Aim : To evaluate the protective function of reducing glutathione ( GSH ) on airway epithelial in guinea pig models of asthma. Methods :48 guinea pigs were randomly divided into 4 groups: the control group, the model group, dexa-methasone group and reduced glutathione group. The asthmatic model was established by ovalbumin challenge method. The concentration of GSH in tissue homogenate of right lung was measured and the left lung was stained with haematoxylin and eosin. The expression of gamma glutamyl cysteine syntase(γ-GCS ) and tumor necrosis factors-a( TNF-α ) was investigated using immunohistochemistry technique. Results : The proportion of epsinophils ( EOS ) and epithelial cells in BALF obtained from reduced glutathione group were statistically higher than that in model group( F = 69. 639 ,62. 306 ,P < 0. 001 ). Compared with model group, the guinea pigs in reduced glutathione group developed less infiltration of inflammatory cells, more integrated epithelial cells and less damage of basement membrane. The concentration of GSH in reduced glutathione group was similar with that in the control group but was higher than that in of model group ( F = 113.047 ,P <0. 001 ). Although immunohislochemistry results indicated that both γ-GCS and TNF-α were observed in bronchial wall and peribronchial distribution, the levels of γ-GCS and TNF-α in reduced glutathione group were significantly lower compared with model group ( F =64.444,89. 895, P < 0.001 ). Conclusion: Reduced glutathione may have the ability to protect airway epithelial cells in guinea pig models of asthma by enhancing oxidation resistance and reducing inflammation.关键词
