Abstract
Aim: To investigate the eilect of mycophenolate mofetil(MMF ) on the expression of insulin-like growth fac-tor-l( IGF-1 ) and matrix metalloproteinase-2( MMP-2 ) in kidney of diabetic rats. Methods : DM rat models induced by in-traperitoneal injection with streptozotocin were constructed. Male SD rats were divided into three groups: normal control rats ( control group ),diabetic rats( DM group ),and diabetic rats received MMF( MMF group ). Eight weeks later,24-hour pro-teinuria quantitation, blood glucose, creatine clearance rate( Ccr ), and the ratio of kidney weight/body weight were measured , and kidney pathology was observed. Immunohistocytochemistry and Western blot were used to analyze the expression of insulin-like growth factor-l( IGF-1 )and matrix metalloproteinase-2( MMP-2 ). Results: The differences on the ratio of kidney weight/body weight, blood glucose, 24-hour proteinuria quantitation and creatine clearance rate( Ccr) among three groups were statistically significant F = 51. 097 ,150. 925 ,161. 315 and 103. 307 ,P < 0. 001 ). The ratio of kidney weight/ body weight,24-hour proteinuria quantitation and creatine clearance rate( Ccr ) of MMF group were lower as compared with DM group and increased markedly as compared with the control group. The PAS staining of sectiones of nephridial tissue showed that the degree oi glomerular basement membrane thickening, mesangium cell hyperplasia, compression of capillary loop and extracellular matrix hyperplasia of the DM group increased significantly as compared with the Control group. Improvements were observed in the MMF group. The differences on the expression of IGF-1 and MMP-2 in renal tissue among three groups were statistically significant(F = 15. 715 and 20. 449 ,P < 0. 001 ). Western blot analysis noted that compared with control group the expression of IGF-1 in renal tissue of DM group was increased. MMF treatment could reduce the increased expression of IGF-1. The expression of MMP-2 was on the contrary. Conclusion : The protective role of MMF in diabetic nephropathy which reduce extracellular matrix deposition in renal tissue possibly associated with the decrease of IGF-1 and the increase of MMP-2.关键词
