Cloning and Bioinformatics Analysis of One Vacuolar H+-Pyrophosphatase Gene in Rehmannia glutinosa, f.hueichingensis (Chan et Sehih) Hsiao

Abstract
In this study,to clone Rehmannia glutinosa functional genes,a pair of degenerate primers was designed using the software CLUSTAL 2. 1 and DNAMAN 4. 0 according to the conserved regions of the five known plant ARGOS base sequences from GenBank nucleic acid database in NCBI. One 495 bp-cDNA fragment of Rehmannia glutinosa. f. hueichingensis (Chan Et Sehih ) Hsiao was cloned using RT-PCR. BLAST analysis indicated that its base sequence was homologous to those of seven known plant vacuolar H+-pyrophosphatase (V-PPase) genes from GenBank with an identity of 83%-86% ,showing that it was V-PPase gene in Rehmannia glutinosa. f. hueichingensis(Chan Et Sehih) Hsiao.关键词
